Triazoles Synthesis & Applications as Nonsteroidal Aromatase Inhibitors for Hormone-Dependent Breast Cancer Treatment
نویسندگان
چکیده
In the last few years, nonsteroidal aromatase inhibitors (AIs) have been emerged as promising agents for treating hormone-dependent breast cancer in postmenopausal women because of their inhibitory effect on estrogen synthesis. Indeed, these compounds can block activity aromatase, enzyme that intervenes steps production pathway. Triazoles are core structures AIs. The nitrogen atom triazole moiety plays a fundamental role functionality by interacting with iron ions heme group. general, AIs possess numerous advantages they quench step synthesis without any effects other steroids produced via same Some anastrozole, letrozole, and vorozole already approved Food Drug Administration treatment cancer. previously mentioned present severe adverse polycystic ovary syndrome (PCOS), resistance onset long-term treatments, higher risk bone fractures. This review focuses intensively hormone-sensitive types cancers, especially triazoles Also, provides an overview about chemistry along different methods which v -triazoles s-triazoles synthesized.

Commentary (Harvey): Nonsteroidal and Steroidal Aromatase Inhibitors in Breast Cancer
Hormone-dependent disease is an important and prevalent subtype of breast cancer. It is recognized by the presence of a functional estrogen-receptor apparatus, an indolent clinical course, a greater frequency in older women, and clinical responses to the sequential use of hormonal therapies. Antiestrogens, primarily tamoxifen (Nolvadex), have been the mainstay of hormonal therapy for metastatic...

Anastrozole (Arimidex), letrozole (Femara), and exemestane (Aromasin) are members of the third generation of aromatase inhibitors that has now replaced aminoglutethimide (Cytadren), the progestins, and tamoxifen (Nolvadex) as the hormonal therapy of choice in estrogen-receptor-positive, postmenopausal, metastatic breast cancer. This article will review the role of aromatase in the pathogenesis ...

The development of aromatase inhibitors for breast cancer therapy is a result of successful translational research exploring the biochemical effects of different compounds in vivo. Studies assessing plasma oestrogen levels as well as in vivo aromatase inhibition have revealed a consistent difference with respect to biochemical efficacy between the third generation compounds (anastrozole, letroz...

متن کاملAromatase inhibitors in the treatment of breast cancer.
A number of inhibitors of estrogen synthesis are now becoming available which could be of value in the treatment of breast cancer. 4-Hydroxyandrostenedione (4-OHA), the first of these compounds to enter the clinic has been found to be effective in postmenopausal patients who have relapsed from tamoxifen. Thus, in studies of 240 patients, 26% patients experienced partial or complete response to ...
